Mitogenic effect of sericin on mammalian cells

چکیده
Fetal bovine serum (FBS) or other mammal-derived factors are extensively supplemented as growth factor into culture medium. Since mammal-derived factors arouses the concern about the risk of zoonosis, serumand mammal-free culture is strongly required. We focused on sericin hydrolysates, originating from silkworm, and reported that sericin is effective as growth factor to various cells. But it is not elucidated how sericin induces the proliferation and inhibits apoptosis of the cells. In this study, inhibitor assay was done in order to identify signaling factors involved in sericin effect. In hybridoma cells, the involvement of Src was suggested, while ERK1/ 2, PP2A and p38 were not involved. In HepG2 cells, the involvement of Src and ERK1/2 was suggested. From these results, it was implied that the mitogenic effect of sericin might be transduced through different pathways depending on cell line.

Sericin is an important peptide derived from silk fibre spun by the silkworm Bombyx mori and has various biological activities. The aim of the present study was to characterize the major constituents of sericin that are providing cytoprotective effect against hydrogen peroxide-induced cell damage in midgut epithelial cells and hemocytes of silkworm. Extracted sericin was subjected to LCMS analy...

Fetal bovine serum (FBS) shows obvious deficiencies in cell culture, such as low batch to batch consistency, adventitious biological contaminant risk, and high cost, which severely limit the development of the cell culture industry. Sericin protein derived from the silkworm cocoon has become increasingly popular due to its diverse and beneficial cell culture characteristics. However, systematic...
متن کاملSilk protein, sericin, suppresses DMBA-TPA-induced mouse skin tumorigenesis by reducing oxidative stress, inflammatory responses and endogenous tumor promoter TNF-alpha.
This study was conducted to assess protective effect of an antioxidant protein, sericin, on tumor promotion in the 7,12-dimethylbenz (alpha) anthracene (DMBA)-initiated and 12-O-tetradecanoylphorbol 13-acetate (TPA)-promoted mouse skin tumorigenesis model. In experiment 1, sericin was applied topically to DMBA-initiated female ICR mouse skin at the doses of 2.5 and 5 mg twice per week for 16 we...
متن کاملThe Effects of Novel Mutations in A1 Domain of Human Coagulation Factor VIII on Its Secretion Level in Cultured Mammalian Cells
Inefficient secretion of the human coagulation factor (hFVIII) in mammalian expression systems is one ofthe main causes of the hFVIII low expression level, attributed to its interaction with a chaperone known asBiP/GRP78. In order to improve secretion efficiency of the hFVIII, based on the higher secretion level of theporcine FVIII and analysis of the hFVIII A110 region, that ...
متن کاملSupplemental Silk Protein , Sericin in 1 , 2 - Dimethylhydrazine - Treated and Cell Proliferation , Suppresses Colon Tumorigenesis Mice by Reducing Oxidative Stress
This study was done to discover tbe underlying mechanism of the inhibitory effect of sericin against colon tumorigenesis. Mice were fed a diet with 30 g!kg sericin for 115 d, and giyen a weekly iajection of 1,2dimethylhydrazine (10 mglkg body weight) for the initial 10 wk. Dietary supplemental sericin caused a 62% reduction in the incictence of colonic adenoma (P<O.05), but did not affect the i...
